Pacquiao had earlier said that he would beat the American for the good of boxing.
The Filipino fighter has a record of 57 wins, two draws and five losses.
"May 2 - the world stops. Mayweather v Pacquiao is the biggest fight in history," Mayweather said.
The fight, which has been the subject of on-off contract negotiations over six years, is expected to generate as much as £162m ($250m).
Who won the verbals?
In low-key speeches in front of about 700 media, both fighters spent more time talking up the fight than aiming barbs at their opponent.
Mayweather, 38, suggested that his unblemished record may give him a psychological advantage over Pacquiao, claiming that "if you lose, it is always in your mind", but also praised the Filipino's camp for making the fight happen.
"I have to be thankful that we have all come together as one to give the world what they want to see," he added.
Pacquiao's confident prediction was retweeted nearly 6,000 times
Pacquiao, who made several references to his Christian faith in an unremarkable address at the news conference, had tweeted criticism of Mayweather's lavish lifestyle earlier in the day.
"Beating Floyd is good for boxing. When athletes have great success, their success goes to their head. That is bad for boxing," he wrote.
"I'm here to prove that I can easily beat the undefeated."
The fight is expected to boil down to whether Pacquiao's speed and power can ruffle Mayweather's supreme defensive skills. Pacquiao was in no doubt he could win, referencing his victories over heavier-hitting fighters than Mayweather.
"My footwork and hand combinations will be my advantage. I tell you Miguel Cotto and Antonio Margarito punch hard. This is boxing and its about punches," he added on Twitter.
"We're gonna kick his ass!"

It was left to Pacquiao's trainer Freddie Roach to deliver the usual pre-fight bombast.
"We're fighting the best fighter in the world - but we're gonna kick his ass," he told the media, drawing smiles from Mayweather.
Earlier, on the red carpet at a Nokia Theatre venue that is more used to hosting the Emmy television awards, he told BBC Sport why he was confident Pacquiao, 36, could inflict Mayweather's first defeat.
"My guy is too fast, he hits too hard. I think Floyd's legs are shot," he said.
He says he wants to stand and trade more and entertain the fans more, but I don't think that is true. He has to say that."
Hottest ticket in town
The cost of seeing the fight in the person was revealed with ticket prices to range from £1,004 ($1,500) to £5,023 ($7,500).

However, it is widely expected that tickets will never actually go on general sale, with celebrities and connections expected to fill the MGM Grand to capacity.
It has been reported that even the well-connected may need to guarantee a certain spend at the venue's casinos to ensure a ringside seat.
Justin Bieber seems likely to be in attendance. The Canadian pop star, a friend of Mayweather's, made a cameo appearance as the fighters posed of more publicity shots.
LOS ANGELES – As the hype machine for boxing's fight of fights cranked into action on Wednesday it was the camp of Manny Pacquiao, and not Floyd Mayweather, who came out firing the first verbal jabs.
It was a role reversal perhaps as surprising as the fact that the clash between the sport's two biggest names is finally taking place, as the typically brash Mayweather stayed subdued while his Filipino opponent and his cohorts let fly.
The day of the only public meeting between the pair ahead of fight week — the contest will take place in Las Vegas' MGM Grand Garden Arena on May 2 — began with Pacquiao, usually quiet and reserved in interviews, taunting Mayweather by stating that the American would lose in a fight with female MMA star Ronda Rousey.
Later, at a scheduled media session, Pacquiao's coach Freddie Roach asserted that Mayweather's legs, at the age of 38, were "shot" and that he has slowed significantly in recent years.
Pacquiao himself said he was less concerned about this fight than he had been before he faced Miguel Cotto, Antonio Margarito or Oscar De La Hoya, all fights he won with consummate ease.
"My feeling is I am confident 110%," Pacquiao said. "I'm not worried about this fight, I was more worried the others."
With less than eight weeks to go until the fight this is usually Mayweather's time to get to work in cutting down the psyche of his opponent. Often his rivals are beaten before they step into the ring, destroyed mentally by a bombardment of taunts and jibes.
It happened with Robert Guerrero, Ricky Hatton, Victor Ortiz and countless others on Mayweather's way to a 47-0 record. Yet this time there is no long, national publicity drive to give Mayweather the daily chance to put doubt into his rival's mind.
The fight was finally inked on February 20 after five years of frustration. With Mayweather set on the May 2 date there was no time for a grand global media tour — Wednesday's press conference, red carpet appearance and media session at Los Angeles' Nokia Theater served as an all-in-one promotional extravaganza.
On his one chance to get inside Pacquiao's head on Wednesday, Mayweather was quiet, reserved and respectful.
"On May 2, I have a tough test," Mayweather said. "Manny Pacquiao is a good fighter. I can't see how it will play out, I am not a psychic. But you best believe I will be in top shape and the best I can be."
It was possibly the clearest signal yet that Mayweather realizes that this is a challenge unlike any he has faced, one that can't be one won with words and actions and publicity, but only with his own mental wiles and silky skills in the ring.
In reality, this is a fight that sells itself, without need for the typical hoopla. Sure, it should probably have happened years ago, but the wait has merely served to stoke the public appetite for what Pacquiao promoter Bob Arum correctly described as the biggest fight of this century, at least to date.
Mayweather went a little further.
"I can remember when I was young and always thought when Marvin Hagler and Sugar Ray Leonard fought in the Superfight, I never thought there would be a fight that was bigger than that. And here we are," Mayweather said.
Both fighters are deeply ensconced in training camp before the bout that is likely to define the legacy of each man. Much will be made of the persona of each man on Wednesday, Mayweather's atypical reticence and Pacquiao's cockiness.
Perhaps Mayweather truly is nervous. Maybe it was a ploy. Maybe Pacquiao is truly relaxed and convinced he can pull off the upset, or maybe his chilled nature was a mask for inner nerves.

"It's been a long road," Mayweather said. "But the fight is on now."
Except that it's not, not quite yet. Until then, every word, every rumor, every nuance will be analyzed with anticipatory fervor.
